Beschreibung Calcitonin (CT) is a 32 amino acid peptide hormone derived from a precursor containing 136 amino acids and an N-terminal leader sequence. CT contains a carboxyterminal proline amide and a disulphide bridge between cysteine residues at position 1 and 7. The sequence is highly conserved in a variety of species within the N-terminal loop region but demonstrates divergence in the rest of the sequence. CT is produced in the parafollicular C-cells of the thyroid and acts through its receptors (CTR) causing osteoclast-mediated bone resorption and calcium excretion by the kidney. Accumulating evidence supports the existence of non-thyroidal CT-like peptides in tissues such as brain, prostate and uterus, which suggest additional physiological roles of CT (2). Calcitonin is a reliable tumour marker as it is secreted by normal C-cells as well as in MTC. MTC may be sporadic ( ∼75% of cases) or familial (∼25% of cases), the latter has an autosomal dominant pattern of inheritance (1, 3).
Spezifität The antibody reacts with human calcitonin. Traces of contaminating antibodies have been removed by solid-phase absorption with human plasma and bovine serum proteins. The antibody labels human thyroid C-cells, and other calcitonin-containing cells strongly and specifically. No non-specific staining has been observed. As demonstrated by immunocytochemistry, the antibody cross-reacts with the calcitonin-equivalent protein in horse and swine.

Anwendungen

Applikationshinweise Polyclonal Rabbit Anti-Human Calcitonin, code No. A 0576, is intended for use in immunocytochemistry. The antibody labels calcitonin-producing C-cells, from which medullary thyroid carcinoma (MTC) originates (1). Differential identification is aided by the results from a panel of antibodies. Interpretation of results must be made within the context of the patient's clinical history and other diagnostic tests by a certified professional. Introduction: The antibody can be used for labelling paraffin-embedded tissue sections fixed in formalin. Pre- treatment of tissues with heat-induced epitope retrieval is recommended. Optimal results are obtained with DakoCytomation Target Retrieval Solution, code No. S 1700, DakoCytomation Target Retrieval Solution, High pH, code No. S 3308, 10 mmol/L citrate buffer, pH 6.0, or 10 mmol/L Tris buffer, 1 mmol/L EDTA, pH 9.0. Pre-treatment of tissues with proteinase K was found inefficient. The tissue sections should not dry out during the treatment or during the following immunocytochemical staining procedure. Dilution: Polyclonal Rabbit Anti-Human Calcitonin, code No. A 0576, may be used at a dilution range of 1:400-1:800 when applied on formalin-fixed, paraffin-embedded sections of human thyroid tissue and using 20 minutes heat-induced epitope retrieval in DakoCytomation Target Retrieval solution, code No. S 1700, and 30 minutes incubation at room temperature with the primary antibody. Optimal conditions may vary depending on specimen and preparation method, and should be determined by each individual laboratory. The recommended negative control is DakoCytomation Rabbit Immunoglobulin Fraction (Solid-Phase Absorbed), code No. X 0936, diluted to the same protein concentration as the primary antibody. Unless the stability in the actual test system has been established, it is recommended to dilute the product immediately before use or dilute in DakoCytomation Antibody Diluent, code No. S 0809. Visualization: DAKO LSAB/HRP kit, code No. K 0679, and DAKO EnVision/HRP kits, code Nos. K 4008 and K 4010, are recommended. Follow the procedure enclosed with the selected visualization kit. Automation: The antibody is well-suited for immunocytochemical staining using automated platforms, such as the DakoCytomation Autostainer. Cells labelled by the antibody display a cytoplasmic staining pattern. Normal tissues: The antibody labels parafollicular C-cells of the thyroid gland. No labelling is observed in the adrenal gland (4). Abnormal tissues: The antibody labels medullary thyroid carcinomas (1). In addition, the antibody labelled 6/28 pheochromocytomas (4), 7/37 carcinoids of the duodenum (5), 2/24 gastric carcinoids (6), and tumour cells in 11/15 cases - and surface cells in 2/15 cases of sclerosing hemangiomas of the lung (7). No labelling was observed in 27 cases of adrenocortical carcinomas (4).

At product concentrations, though not classified as hazardous, sodium azide may react with lead and copper plumbing to form highly explosive build-ups of metal azides. Upon disposal, flush with large volumes of water to prevent metal azide build-up in plumbing. 3. As with any product derived from biological sources, proper handling procedures should be used. 4. The product may be used in different techniques and in combination with different sample types and materials, therefore each individual laboratory should validate the test system applied.
