Glucagon, a polypeptide of 29 amino acids, is one of seven known hormones produced in the pancreas. Glucagon, secreted by A-cells of the islets of Langerhans, elicits elevated blood glucose concentrations, stimulates intestinal fluid secretion and gastrin release and facilitates the inhibition of gastric and pancreatic secretions. 1,2

Spezifität
|
The antibody has not been characterized by RIA techniques. Cross-reactivity with glucagon from other mammalian species was demonstrated by immunohistochemistry. This product has been optimized for use on human tissue.

Applikationshinweise
|
Paraffin Sections Rabbit anti-glucagon can be used on formalin-fixed, paraffin-embedded tissue sections. Pretreatment of tissue with proteolytic enzymes is not required. Follow the recommended procedure for the detection system selected. Staining interpretation The cellular staining pattern for anti-glucagon is cytoplasmic. Normal Tissues This antibody labels the cytoplasm of glucagon-producing A-cells. A-cells found on the periphery of pancreatic islets, constitute approximately 20% of normal islet cell populations. A-cells are also present in extrainsular locations such as along the pancreatic ductal epithelium. 3 (107192-001) 303256EFG_001 p. 2/4 Abnormal Tissues Tumors of the pancreatic islet cells are frequently multihormonal, and their classification is based upon the predominant hormone present. Glucagonomas predominantly produce glucagon or its precursors and constitute approximately 8% of all pancreatic endocrine tumors.

Rabbit anti-glucagon is an unfractionated rabbit antiserum in 0.05 mol/L Tris-HCL, pH 7.6, and 0.015 mol/L sodium azide.

Store at 2-8 °C. Precautions: 1. For professional users. 2. This product contains sodium azide (NaN 3 ), a chemical highly toxic in pure form. At product concentrations, though not classified as hazardous, NaN 3 may react with lead and copper plumbing to form highly explosive build-ups of metal azides. Upon disposal, flush with large volumes of water to prevent metal azide build-up in plumbing. 3. As with any product derived from biological sources, proper handling procedures should be used. 4. Wear appropriate Personal Protective Equipment to avoid contact with eyes and skin. 5. Unused reagents should be disposed of according to local, State, and Federal regulations.
