    Western Blotting (WB), Immunohistochemistry (IHC)
    Spezifität
The antibody detects endogenous level of HDAC5 only when phosphorylated at serine498.
    Aufreinigung
The antibody was affinity-purified from rabbit antiserum by affinity-chromatography usingepitope-specific phosphopeptide. The antibody against non-phosphopeptide was removedby chromatogramphy using non-phosphopeptide corresponding to the phosphorylation site.
    Immunogen
Peptide sequence around phosphorylation site of pSer498 (T-Q-S (p) -S-P) derived from Human HDAC5, 7. Antibodies were produced by immunizing rabbits with synthetic phosphopeptide and KLH conjugates.

    Hintergrund
Histones play a critical role in transcriptional regulation, cell cycle progression, and developmental events. Histone acetylation/deacetylation alters chromosome structure and affects transcription factor access to DNA. The protein encoded by HDAC5 belongs to the class II histone deacetylase/acuc/apha family. It possesses histone deacetylase activity and represses transcription when tethered to a promoter. It coimmunoprecipitates only with HDAC3 family member and might form multicomplex proteins. It also interacts with myocyte enhancer factor-2 (MEF2) proteins, resulting in repression of MEF2-dependent genes. This gene is thought to be associated with colon cancer. Two transcript variants encoding different isoforms have been found for this gene. Doppler H, et al. (2005) J Biol Chem. 280(15):15013-15019. McKinsey TA, et al.
