Beschreibung The Grb2-associated binder 1 (Gab1) is a multisubstrate scaffold/adapter protein thatcontains a pleckstrin homology domain at the NH2 terminus and multiple tyrosyl residuesand proline-rich motifs for association with src homology 2 (SH2)-containing andSH3-containing proteins. Gab1, as well as its close relatives Gab2 and Gab3, serves tocouple signaling between cell surface receptors for growth factors and cytokines with avariety of downstream targets, including Shp2 tyrosine phosphatase, phosphatidylinositol3-kinase (PI3K), and phospholipase C-gamma. Ablation of the Gab1 gene in mice resultedin embryonic death in homozygous mutants, with developmental defects in the placenta,heart, and skin. Biochemical analysis of Gab1-/- fibroblast cells suggested a positive roleof Gab1 in coupling the signal relay between cytokine receptors and the ERK pathway.
