SLC7A7, also called y + L-type amino acid transporter 1 (y + LAT1), forms a heterodimeric complex with the heavy chain of the cell surface antigen 4F2 (4F2hc/CD98hc/SLC3A2). SLC7A7 is mainly expressed at the basolateral membrane of epithelial cells in the renal tubules and small intestine, and transports cationic amino acids such as lysine, arginine, and ornithine. Functionally deficient variants of the SLC7A7 gene have been observed in patients with lysinuric protein intolerance, which is characterized by increased urinary excretion of cationic amino acids and reduced absorption from intestine, incidentally causing metabolic derangement after protein-rich meals. Western blotting, 1:1,000 for chemiluminescence detection system Immunoprecipitation, Not tested Immunohistochemistry, 1:1,000 Heat treatment is necessary for staining paraffin embedded sections. Autoclave, 125 o C for 5 minutes in 10 mM citrate buffer containing 0.05% Tween-20 (pH 6.0). Immunocytochemistry, Not tested Flow cytometry, Not tested . Protocol: Immunohistochemical staining for paraffin-embedded sections 1) Deparaffinize the sections with Xylene 3 times for 3-5 minutes each. 2) Wash the slides with Ethanol 3 times for 3-5 minutes each. 3) Wash the slides with PBS 3 tim Heat treatment es for 3-5 minutes each. 4) Heat treatment by Autoclave: Heat the slides immersed in retrieval solution [10 mM citrate buffer containing 0.05% Tween-20 (pH 6.0)] at 125oC for 5 minutes in pressure boiler. After boiling, the slides should remain in the pressure boiler until the Immunohistochemical detection of SLC7A7 on paraffin embedded section of human kidney with BMP019. Tissue array (MBL, code no.
