Beschreibung
|
Inhibitor of NFkappaB alpha (IkappaBalpha) is a ~40 kDa protein that has a central role in the regulation of transcription mediated by the NFkappaB transcription factor. In the absence of a stimulus, NFkappaB is sequestered in the cytoplasm through binding to IkappaB proteins. A variety of stimuli (e.g., proinflammatory cytokines, Tumor Necrosis Factor- alpha, and interleukin-1beta) lead to phosphorylation and activation of the IKK complex (consisting of IKK alpha,beta, and gamma). The activated IKK complex in turn phosphorylates IB alpha serines 32 and 36, leading to its subsequent ubiquitination and degradation by the proteasome. The free NFkappaB translocates to the nucleus and stimulates transcription of NFkappaB-responsive genes. Phosphorylation of these serine residues by several other kinases, including p90 S6 kinase (Rsk1), NFkappaB-Inducing Kinase (NIK), and Mitogen-Activated Protein Kinase Kinase Kinase-1 (MEKK1), with subsequent activation of NFkappaB-mediated transcription, has also been reported. A component of the NFkappaB complex, RelA, has recently been shown to regulate the level of IkappaB alpha phosphorylation.

Human I kappaBalpha. Mouse and pig (100% homologous), rat and chicken (93% homologous) I kappaBalpha have not been tested, but are expected to react. APPLICATIONS : The antibody has been used in Western blotting. For Western blotting applications, we recommend using the antibody at a 1:1000 starting dilution. The optimal antibody concentration should be determined empirically for each specific application

Purified from rabbit serum by sequential epitope-specific chromatography. The antibody has been negatively pre-adsorbed using a non-phosphopeptide corresponding to the site of phosphorylation to remove antibody that is reactive with non-phosphorylated I kappaBalpha. The final product is generated by affinity chromatography using an I kappaBalpha-derived peptide that is phosphorylated at serines 32 and 36.
