Macrophages and histiocytes, which are widely distributed throughout the body, play acrucial role in the body defense mechanisms. They are heterogenous in morphology,cytochemistry, function and surface antigens. Immunohistochemical labeling withmonoclonal antibodies is one of the most specific and reliable techniques for theidentification and localization of these cells. Monoclonal antibodies to macrophage andhistiocyte surface or cytoplasmic antigens are important for the immunodiagnosis ofboth benign and malignant histiocytic lesions. They are useful in the investigation ofmacrophage differentiation and maturation and in the diagnosis of various inflammatory,reactive and neoplastic disorders of macrophages. Spezifität

The antibody recognizes an antigen present in the cytoplasm of human macrophages ina wide variety of tissues and organs. The epitope recognized is resistant toformalin-fixation and paraffin-embedding. This antibody may also be used on materialfixed with B5 solution, neutral buffered formalin, zinc-formalin, Brunnel',s, Susaand Bouin',s solutions but not in standard frozen sections. The antibody stains thecytoplasm of macrophages and histiocytes in hematopoietic organs, Kupffer',scells of the liver, and Langerhans', cells of the skin. Mantle zone B lymphocytesand interdigitating reticulum cells of lymph node and B cells in spleen are also stained.Bone marrow lymphoid and myeloid precursor cells stain with a uniform interstitialpattern. Certain normal non-lymphoid cells such as spermatogonia are stained whileothers such as breast ductal epithelium, kidney tubular epithelium, endometrium,parathyroid and stomach chief cells are weakly stained. The antibody is reactive withbenign and malignant histiocytic lesions such as sinus histiocytosis, dermatopathiclymphadenopathy, persistent generalized lymphadenopathy, tuberculosis, sarcoidosis,Gaucher',s disease, true histiocytic lymphomas, malignant histiocytosis andhistiocytosis-X. Species Reactivity: Cross-reacts with Human.Not yet tested in other species.

IF: Use at a dilution of 1/400. The epitope recognized by the product is resistant toformalin fixation and paraffin embedding. The antibody also may be used on materialfixed with B5 solution, neutral buffered formalin, Zinc-Formalin, Brunnel',s, Susaand Bouin',s solutions. The antigen is not detectable by the antibody in standardfrozen sections. Using indirect immunofluorescent or immunoperoxidase methods, theantibody stains the cytoplasm of macrophages and histiocytes in hematopoietic organsand Kupffer cells of the liver and Langerhans cells of the skin. Mantle zone Blymphocytes, interdigitating reticulum cells of lymph nodes and B cells in spleen arestained. Bone marrow lymphoid and myeloid precursor cells stain with a uniforminterstitial pattern.
