Beschreibung
|
Apoptosis plays a major role in normal organism development, tissue homeostasis, andremoval of damaged cells. Hrk, a pro-apoptotic member of the Bcl-2 homology domain-3(BH3) group of the Bcl-2 family of proteins, was identified as a novel protein inducedduring programmed neuronal death. It lacks significant homology to other Bcl-2 familymembers except for an 8-amino acid region that is similar to the BH3 motif of Bik. Hrkregulates apoptosis through interaction with the anti-apoptotic proteins Bcl-2 and Bcl-XLvia this domain. It does not interact with the pro-apoptotic proteins Bax, Bak, or Bcl-XS.Hrk localizes to mitochondrial membranes in a pattern similar to that previously reportedfor Bcl-2 and Bcl-XL. Despite its predicted molecular weight, Hrk often migrates at 12-15kDa. ABIN260674 detects Harakiri (HRK), also known as DP5. HRK is a BH3 only protein ofthe Bcl-2 family, which promotes Bax dependent apoptosis. HRK regulates apoptosisthrough its interaction with the anti-apoptotic proteins Bcl-2 and Bcl-XL, and inconjunction to the mitochondrial pore forming protein p32. HRK is thought to have aspecific role in initiating neuronal and haematopoietic cell apoptosis, in particular togrowth factor deprivation.Aberrant methylation of HRK is frequently detected in primary colorectal cancers.Despite its predicted molecular weight, HRK often migrates at 12-15kD.
