Beschreibung: The receptors on T cells consist of immunoglobulin like integral membrane glycoproteinscontaining 2 polypeptide subunits, alpha and beta, of similar molecular weight, 40 to 55kD in the human. Like the immunoglobulins of the B cells, each T cell receptor subunithas, external to the cell membrane, an N terminal variable domain and a C terminalconstant domain. T cell receptors recognize foreign antigens which have beenprocessed as small peptides and bound to major histocompatibility complex moleculesat the surface of antigen presenting cells. Each T cell receptor is a dimer consisting ofone alpha and one beta chain or one delta and one gamma chain. In a single cell, the Tcell receptor loci are rearranged and expressed in the order delta, gamma, beta, andalpha. If both delta and gamma rearrangements produce functional chains, the cellexpresses delta and gamma. If not, the cell proceeds to rearrange the beta and alphaloci.

Spezifität: Detects a minor subset of T cells. The subpopulation of HIS 42 positive T cells containsboth CD4 and CD8 positive cells, in the same ratio as found in the peripheral T cellpopulation. When bound to Sepharose beads, HIS 42 induces T cell proliferation in thepresence of IL-2. In contrast to lymph node T cells a number of thymocytes are found toexpress HIS 42 only in the cytoplasm or together with membrane expression. Mostbright HIS 42 surface labeled thymocytes are also positive for MRC OX-44, a markerpredominantly identifying mature thymocytes. SDS PAGE analysis of the membranemolecules immunoprecipitated by HIS 42 shows a band at 85kD. On reduction itseparates to two bands at 35kD and 48kD. From this data it is concluded that HIS 42 isdirected against a determinant on the rat T cell receptor, which is common to a smallnumber of T cells. Preliminary data suggests that the determinant is found on the betachain, associated with the Vbeta16.1 gene product. Species Reactivity: Reacts with Rat.
