Beschreibung: ADAM22 was first described as MDC2 (Metalloproteinase-like disintergin like cysteinerich 2) from analysis of human brain libraries, in search of brain-specific proteins. Twosplice variants with different carboxyterminal ends have been described. ADAM22participates in cell adhesion and spreading by associating with 14-3-3 proteins. It isconstitutively produced in normal brain tissue, but decreased or absent in high gradegliomas. Different splice variants of ADAM22 are differentially expressed in areas of thebrain. A complex of ADAM22 and LGI1 were shown to regulate synaptic transmission.Complexes of ADAM22, stargazing, and a range of other proteins anchor ADAM22 intothe cytoskeleton, and keep ADAM22 in association with the postsynaptic space.ADAM22 does not contain the canonical HExxHxxxxxH zinc metalloproteinase motif,and is not thought to be proteolytically active. ADAM22 is a member of the ADAMS thatfunction in tandem with other ADAMs and which may also have specific individualfunctions.
