Beschreibung: Rabbit anti Thymidylate Synthase Rabbit Antibody to Human Thymidylate Synthase

Spezifität: Detects Human Thymidylate Synthase (36kDa). Also recognizes an additional band corresponding to the ternary complex of human TS-dFUMP-reduced folate in HeLa cells treated with the TS inhibitor 5-FUdR. This antibody can detect antigen in a variety of human cells and tissues, as well as bacteria, African green monkey, rat and mouse. Can detect Thymidylate Synthase by immunochemistry in proliferating cell cultures and tissues but does not stain nonproliferating cells. Normal colon mucosa shows weak staining, however, some colorectal cancer specimens show very strong staining.

Anwendungen

Applikationshinweise: Assayed by immunoblot using HRP Goat anti Rabbit IgG (H&L) and TMB as a substrate. A working dilution of 1:1,000 is suggested for immunoblotting. A methanol based fixative can be used for immunohistochemistry using a 1:100 working dilution of the antiserum. To immunoprecipitate antigen a dilution of 1:500 is suggested. For immunoprecipitation, pre-clearing with a non-specific rabbit IgG is helpful to reduce background. For ELISA a working dilution of 1:100,000 is suggested using HRP Conjugated Goat anti Rabbit IgG (H&L) and ABTS as a substrate for 30 minutes at room temperature. Each laboratory should determine an optimum working titer for use in its particular application. Other applications have not been tested but use in such assays should not necessarily be excluded.

Lagerung: Aliquot and store at -20°C. Avoid multiple freeze/thaw cycles. Prepare working dilution only prior to immediate use. : This product contains sodium azide, which has been classified as Xn (Harmful) in European Directive 67/548/EEC in the concentration range of 0.1-1.0%. When disposing of this reagent through lead or copper plumbing, flush with copious volumes of water to prevent azide build-up in drains.
