Beschreibung: Amyloid is derived by the sequential cleavage of amyloid precursor protein (APP) by beta- and gamma-secretases. A double missense mutation (Lys670Asn and Met671Leu) in APP found in a Swedish pedigree (APP-sw) elevates Abeta40 and Abeta42 production (1), and the mutation is utilized in establishment of transgenic mice overexpress a mutant form of human amyloid precursor protein (2). Amyloidproduction and, beta-secretase cleavage of APP -sw reportedly occur in the endoplasmic reticulum (ER), Golgi and endocytic compartments (3).

Spezifität: The antibody is specific for soluble Amyloid Precursor Protein of Swedish type cleaved by beta-secretase. No cross reactivities with sAPPbeta-sw of wild type and the full length APP of both Swedish and wild types.

Anwendungen

Applikationshinweise: This antibody can be stained in sAPP-sw gene transfected COS cells after fixation by 20% formalin containing methanol solution without any pretreatment by several cytochemical techniques such as Avidin Biotin Complex (ABC) Method. On the other hand, the antibody can not stained sAPP-wild type gene transfected COS cells by same procedure. The optimal dilution is about 5 ug/ml, however, the dilution rate should be optimized by each laboratories. This antibody can be used for western blotting in concentration of about 5 ug /ml.
