Beschreibung NF-H is an abundant, stable cytoplasmic protein located in neuronal cells in large axons frequently used as a cell type marker. The NF-H protein shares a high degree of structural and sequence homology with the NF-L and NF-M subunits, especially in the coiled-coil core domain. NF-M and NF-H form flexible extensions linking the neurofilament proteins to each other and other cytoplasmic proteins. Deletions and insertions in the NF-H protein have been reported in amyotrophic lateral sclerosis. The NF-01 monoclonal antibody reacts with a conserved phosphorylated epitope of NF-H present in all species and has been reported to be useful for Western blotting, immunohistochemistry using formalin-fixed, paraffin-embedded tissues and immunofluorescence staining. Distribution: Cytoplasmic protein, neuronal cells (large axons) Function: Provides flexible extension (along with NF-M) to link neurofilaments to each other and to other cytoplasmic proteins Modifications: Phosphorylation Synonyms: Neurofilament protein, heavy polypeptide, neurofilament heavy polypeptide Structure: Contains four coiled-coil core domains, in most vertebrates NF-H forms interacts with NF-M (medium chains) and NF-L (light chains), approximately 210 kD. NF-H contains a repetitive sequence motif (Lys-Ser-Pro) repeated more than 40 times.

Anwendungen

Applikationshinweise Additional reported applications (for the relevant formats) include: immunohistochemistry of formalin-fixed, paraffin-embedded tissue sections, immunocytochemistry. Each lot of this antibody is quality control tested by Western blotting. For Western blotting, suggested working dilution(s): Use 5 µg per 5 ml antibody dilution buffer for each mini-gel. It is recommended that the reagent be titrated for optimal performance for each application.
Reinigung The antibody was purified by affinity chromatography.
Buffer This antibody is provided in phosphate-buffered solution, pH 7.2, containing 0.09% sodium azide at 0.5 mg/ml.
