Immunogen: This antibody was purified from hybridoma (clone 5D4) supernatant using protein A Sepharose. This hybridoma was established by fusion of mouse myeloma cell PAI with Balb/c splenocyte immunized with human PRAD1/cyclin D1 gene product from E. coli.

Beschreibung: Cyclin D1, also known as Bcl-1, CCND1, or PRAD-1, is a 36 kDa nuclear protein that plays a key regulatory role during the G1 phase of the cell cycle by regulating the activity of Cdk proteins. Cyclin D1 forms a complex with Cdk4, then binds to and phosphorylates Rb protein, triggering cells to progress from G0/G1 to S and thus driving cellular proliferation. Cyclin D1 protein overexpression is found in the majority of human breast cancers and is usually linked with poor prognosis. Overexpression of cyclin D1 is a well-established criterion for the diagnosis of Mantle Cell Lymphoma, and also has been used to distinguish malignant breast carcinomas from premalignant breast lesions.

Spezifität: This antibody reacts with human cyclin D1 and D2 and mouse cyclin D1 and D2 on Western blot. The antibody does not recognize human cyclin D3 or mouse cyclin D3.

Anwendungen

Applikationshinweise: Western blotting: 1-10 µg/mL. Immunohistochemistry: 1-10 µg/mL Heat treatment is necessary for paraffin embedded sections. Microwave oven: 2 times for 10 minutes each in citrate buffer (pH 6.5). Flow Cytometry: 10 µg/mL (final concentration). Immunoprecipitation: 1-10 µg / 200 µg of cell extract from 5 x 10 6 cells. Immunocytochemistry: not recommended.

Konzentration: 1 mg/mL

Buffer: 100 µg IgG in 100 µL P BS containing 50% glycerol, pH 7.2. Contains no preservatives.
