Beschreibung The two major cytoskeletal proteins implicated in cell motility are actin and myosin. Actinand myosin are constituents of many cell types and are involved in a myriad of cellularprocesses including locomotion, secretion, cytoplasmic streaming, phagocytosis andcytokinesis. Although actin is one of the most conserved eukaryotic proteins, it is expressed in mammals and birds as at least six isoforms characterized by electrophoresisand amino acid sequence analysis. Four of them represent the differentiation markers ofmuscle tissues and two are found in practically all cells.
Spezifität Specific for skeletal and non-muscle actin isoforms (Mr 43,000). Reactivities on Cultured Cell Lines (tested so far): PtK2, SR-NRK, NRK-49F, L6 cells, ratfibroblasts, NIH-3T3, mouse myoblast and myotube cells. Species: Human, Bovine, Rabbit, Chicken and Xenopus laevis. Others not tested.

Anwendungen

Applikationshinweise Immunoblotting. Cytological material. Immunofluorescence Microscopy (Only after methanol fixation). Immunohistochemistry on Frozen and Paraffin Sections: Ready to use forimmunohistochemistry. Incubation Time: 1h at RT (extended with paraffin). With paraffin embedded sections,protease pretreatment is required prior to antibody application. When using antibody 2G2 in immunolocalization studies the presence of nuclear actincould be demonstrated e. g. in cultured cells after fixation with 4% paraformaldehyde. Immunostaining with cytoplasmic actin was only positive after fixation with methanol (cf. Gansior et al. , 1999). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Lagerung Store the antibody undiluted at 2-8°C. Shelf life: one year from despatch.
