Immunogen: A synthetic peptide derived from 154-167 amino acid of human MDM2 protein.

Format: Ig-fraction

Spezifität: Human homology of murine double minute-2 (MDM2) and p53 function in mutual regulation: activation of p53 results in expression of MDM2 which consequently inhibits the ability of wild type p53 to stimulate expression of target genes. MDM2 is frequently overexpressed in the nuclei of soft tissue sarcomas, and to a much lesser extent in a wide variety of malignancies of bladder, breast, testes, cervix, and glial cells. This antibody cross reacts with human, rat and mouse.

Anwendungen

Applikationshinweise: Titer/Working Dilution: This antibody may be diluted to a titer of 1:50-1:100 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. Staining Protocol: We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation conditions should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Positive Control: Breast carcinoma. Buffer: Purified immunoglobulin fraction of rabbit antiserum against MDM2 protein containing sodium azide as preservative.
