Beschreibung: Epitope tagging is a powerful and versatile strategy for detecting and purifying proteinsexpressed by cloned genes. To utilize this feature, protein expression vectors are typicallyengineered with a nucleotide sequence that encodes the peptide epitope tag. The gene ofinterest is cloned in-frame relative to the tag and, upon expression, the protein of interestis synthesized as a fusion protein with the peptide tag. Fusion protein detection and/orpurification is mediated by highly specific antibodies to the engineered peptide, thuseliminating the need for antibodies to proteins from each newly cloned gene. Commonlyused epitope tags include glutathione-S-transferase (GST), 6-histidine (6X-His), greenfluorescent protein (GFP), c-myc and others. The c-myc oncogene (p62 c-myc) is involved in the control of normal cellular proliferationand differentiation. In addition, deregulated expression of c-Myc induces apoptosis indifferent cell types, with c-myc requiring p53 for apoptosis in many cell types. This factindicates heterogeneous mechanisms for c-myc-induced apoptosis.
