Beschreibung
|
Monoclonal antibody X-14, together with X-4, and PM-2K, forms a particular group ofmacrophage specific antibodies which were tested at the Vth Leukocyte Typing Workshopheld in Boston in 1993. The X-14 antigen was initially suspected to function as a scavengerreceptor.

Spezifität
|
Human macrophages, also positive on monkey, weak on rabbit, goat, bovine and horse. Negative on rat, guinea pig, cat, and dog. The antigen is found on the cytoplasmicmembranes of macrophages. Experiments with CHO cells expressing Macrophage ScavengerReceptor (MSR) AI or AII showed that they were not recognized by this antibody.

Protokoll
|
Protocol with frozen, ice-cold acetone-fixed sections: The whole procedure is performed at room temperature1. Wash in PBS2. Block endogenous peroxidase3. Wash in PBS4. Block with 10% normal goat serum in PBS for 30min. in a humid chamber5. Incubate with primary antibody (dilution see datasheet) for 1h in a humid chamber6. Wash in PBS7. Incubate with secondary antibody (peroxidase-conjugated goat anti mouse IgG+IgM(H+L) minimal-cross reaction to human) for 1h in a humid chamber8. Wash in PBS9. Incubate with AEC substrate (3-amino-9-ethylcarbazol) for 12min. 10. Wash in PBS11. Counterstain with Mayer's hemalum.

Applikationshinweise
|
Suitable for Immunohistochemistry on frozen sections: 2 ug/ml (1/100). Suggested positive control: Human tonsil. Antigen distribution on Isolated cells and Tissue sections: Positive on peritoneal macrophages and on cultivated macrophages with serumsupplementation. Positive in AML but not on blast cells. Negative under serum-free conditions, on freshly isolated monocytes and on fMLPactivated neutrophils. U937 cells show increased antigen expression after TNFα and GM-CSF treatment. X-14 is positive on macrophages, Kupffer cells, mesangial cells and Hoffbauer cells. It ispositive on intrafollicular and paracortical dendritic and spindel macrophages and withmacrophages and foamy cells in Xathoma disseminatum but negative in sarcoidgranuloma and histiocytosis X. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
